Chromatographic studies were carried out on aqueous leaves extract of Ventilago maderspatana Gaertn belonging to the family Rhamnaceae. It is an important and well known plant in the Indian system of medicine. Leaf samples of V. maderspatana were subjected to maceration by using distilled water. Aqueous extract were prepared from the leaf, of Ventilago maderspatana. Preliminary phytochemical analysis was carried for the extract. Extract was found to contain different phytoconstituents such as alkaloids, glycosides, steroids and sterols, saponins, Flavonoids, carbohydrates and tri-terpenoids and showed the absence of amino acids, proteins and acidic compounds. Fractions were prepared according to polarity and subjected to TLC and HPTLC analysis....
The simple, rapid, sensitive, precise and specific high-performance thin layer chromatographic (HPTLC) method for the determination of teneligliptin hydrobromide in bulk drug and pharmaceutical dosage form was developed and validated. The concentration range of 500-3000 ng/spot with coefficient of correlation (r2) of 0. 987. For HPTLC the method employed aluminium plates precoated with silica gel G60 F254 as the stationary phase. The solvent system consisted of the chloroform:methanol:glacial acetic acid in the proportion of 8.5:1.5:0.05, v/v/v. This solvent system was found to give compact spots for the teneligliptin HBr with Rf value 0.41 ± 0.01. Densitometric analysis of the teneligliptin HBr was carried out in the absorbance mode at 254 nm. The developed method was validated as per the ICH guidelines. Statistical analysis proved that method is repeatable and specific for the estimation of the said drug. These methods can be adopted in routine assay analysis of the teneligliptin HBr in bulk or tablet dosage form....
Gemcitabine hydrochloride is used as an anti-cancer drug. The aim of this study was to develop colorimetric method for the estimation of gemcitabine in bulk and lyophilized powder for injection. In colorimetric method the absorption maximum was found to be 556 nm and beer’s law obeyed in the concentration range of 10-30 µg/ml. The LOD and LOQ were found to be 0.27 and 0.84 µg/ml respectively with coefficient of correlation 0.997. Molar absorptivity was found to be 7.9 x 105, indicating that the intensity of absorption at the selected absorption maximum is strongest. The method validated for linearity, accuracy and precision as per ICH (Q2, R1) guidelines and applied for the quantitative analysis of gemcitabine hydrochloride in bulk and dosage form....
Uncomplicated, accurate and short analysis time analysis was elaborated for quantification of Articaine HCl (ART HCl) and Epinephrine (EPI). The electrochemical behavior of both articaine and epinephrine was carried out by applying differential pulse voltammetry (DPV) using Pencil graphite (PGE), carbon paste (CPE) and Nanomodified zirconium (ZRE) electrodes. The effect of experimental parameters has been investigated and optimized. The peak current linearity range were 6.62 × 10-5 - 50.60 × 10-5 mol L-1 for ART and 9.90 ×10-5 - 29.1 ×10-5 mol L-1 for EPI using (PGE), 3.90 × 10-5 -15.7× 10-5 mol L-1 for ART 4.97× 10-5 - 17.7 x10-5 mol L-1 for EPI, using (CPE) 5.96 × 10-5 - 23.4x10-5 mol L-1 for ART and 2.99×10-5- 22.5 × 10-5 mol L-1 for EPI using (ZRE). The suggested analysis has been applied for assay of ART and EPI in bulk, pharmaceuticals as well as in healthy human urine with good analytical results. A simple, rapid, economical, precise and accurate reverse phase high performance liquid chromatography (RP-HPLC) method for estimation of doxophylline and acebrophylline in bulk and tablet dosage form had been developed. The separation was attained by Column Hypersil BDS C18, 150 × 4.6 mm, 5 µ and 10mM dipotassium hydrogen phosphate + 0.1% TEA (pH 5.0): Acetonitrile (50:50 v/v) as mobile phase at a flow rate 1.0 ml per minute. Detection was carried out of wavelength of 271 nm retention time of doxophylline were found to be 4.293 min and acebrophylline were found to be 5.526 min to be respectively. The method has been validated for linearity, accuracy and precision. Linearity observed for range of 16.0 to 56.0 μg/ml for doxophylline and 4.0 to 14.0 μg/ml for acebrophylline. The percentage recovery obtained for doxophylline were found to be in range 99.09 to 100.05 and acebrophylline were found to be in range 99.06 to 100.00, respectively. Development method was found to be accurate, precise and rapid for estimation of doxophylline and acebrophylline in bulk and combined dosage form....
Stability indicating RP-HPLC method for simultaneous estimation of clotrimazole, clindamycin phosphate and tinidazole in their combined dosage form has been developed. The separation was achieved by Shimadzu LC-10AT Hypersil BDS C18 column and buffer (pH 4.5): acetonitrile (60:40) as mobile phase, at a flow rate of 1 ml/min. Detection was carried out at 230 nm. Retention time of clotrimazole, tinidazole and clindamycin phosphate was found to be 3.103 min, 4.277 min and 5.707 min, respectively. The method has been validated for linearity, accuracy and precision. Linearity observed for clotrimazole 25-75 microgram/ml, tinidazole 25-75 microgramg/ml and for clindamycin phosphate 25-75 microgramg/ml. Developed method was found to be accurate, precise and rapid for simultaneous estimation of clotrimazole, clindamycin phosphate and tinidazole in their combined dosage form. The drug was subjected to stress condition of hydrolysis, oxidation, photolysis and Thermal degradation. Considerable degradation was found in alkaline degradation. The proposed method was successfully applied for the simultaneous estimation of the drugs in commercial dosage form....
The proposed study described the development of two simple, accurate, cost effective and rapid zero order uv-spectrophotometric-AUC method and high-performance thin layer chromatographic (HPTLC) method for the estimation of remogliflozin etabonate in bulk and in-house Tablets. Remogliflozin etabonate showed maximum absorbance at 225 nm in methanol. For UV-AUC method wavelengths 216.80 nm and 232 nm selected for integration of AUC. The linearity range for zero order UV-AUC was 4-24 microgram/ml and for HPTLC 1500-9000 ng/band. The percent recovery for zero order UV-AUC and HPTLC were found for both the methods in between 98-100%. Both methods were validated according to ICH guidelines. Both depicted methods can routinely be used for the estimation of remogliflozin etabonate in bulk and in-house tablets....
A rapid, simple, selective and precise UV-visible spectrophotometric method has been used for the estimation of caffeine content in various nine brands of tea powder consumed in Nepal. Numbers of reports are available for the estimation of caffeine in both official and published research papers. In Nepal, majority of the people consumed tea rather than coffee. Their daily life starts with a cup of tea. During our study we have found that no any specific research has been done about this chemical constituent on various brands of tea available in the market of Nepal. The amount of caffeine in tea brands were estimated by comparing the absorbance with reference caffeine using multiple standard method. The linearity range was found to be 2-12 μg/ml with a correlation coefficient of 0.998. From our study, the highest amount of caffeine content was found in Nepalese tea, Tokla (6.6212 μg/ml) as compared to other brands of tea such as Indian tea, Taj mahal (5.3939 μg/ml), Red label (5.4393 μg/ml) and Tata 5.8181(μg/ml) and Chinese tea Jasmine (5.4090 μg/ml). Thin layer chromatography was performed to ensure the separation of the spots. Our study may be the first in such setting in Nepal to assess this chemical constituent in different brands of tea....
